Details about Double Stranded DNA Ab
What is the Double Stranded DNA Antibodies Test?
The Double Stranded DNA (dsDNA) Antibodies test measures autoantibodies directed against the body’s own double stranded DNA. These antibodies are highly specific for systemic lupus erythematosus (SLE) and reflect abnormal immune system activity.
Unlike some other autoimmune markers, dsDNA antibodies are closely associated with disease activity in lupus. Levels may rise during disease flares and fall during remission, making this test useful not only for diagnosis but also for ongoing monitoring.
The test is commonly requested following a positive antinuclear antibody (ANA) result or when lupus is clinically suspected.
Why do I need a test for Double Stranded DNA Antibodies?
You may need this test if symptoms suggest an autoimmune condition, particularly lupus. Because lupus can affect multiple organs, early and accurate diagnosis is essential for preventing long-term damage.
Monitoring dsDNA antibody levels also helps assess disease activity and response to treatment in individuals with established lupus.
Symptoms and scenarios where testing is recommended:
- Persistent joint pain or swelling
- Fatigue not explained by lifestyle or illness
- Skin rashes, including photosensitivity
- Mouth or nasal ulcers
- Unexplained fevers
- Kidney involvement or abnormal urine tests
- Suspected or confirmed lupus
- Monitoring autoimmune disease activity
Benefits of the Double Stranded DNA Antibodies Test
- Highly specific for lupus
One of the most specific markers for SLE. - Supports early diagnosis
Helps confirm autoimmune disease when symptoms are unclear. - Monitors disease activity
Levels often correlate with lupus flares. - Assesses kidney involvement risk
High levels are linked to lupus nephritis. - Guides treatment decisions
Helps clinicians adjust therapy appropriately. - Complements ANA testing
Provides greater diagnostic clarity. - Useful for long-term monitoring
Tracks changes over time. - Simple blood-based test
Requires only a standard blood sample.
Step-by-Step: How Testing Works
Choose your preferred testing method
You can attend one of our UK partner clinics or arrange a home or hotel visit with a mobile phlebotomist.
Option 1 – In-Clinic Blood Draw
- Select your nearest UK clinic and book an appointment.
- A clinician or phlebotomist collects a venous blood sample.
- The sample is analysed for dsDNA antibodies.
- Results are delivered securely online.
Option 2 – Home or Hotel Phlebotomy (+£60)
- Book a UK-wide home or hotel visit at a convenient time.
- A trained phlebotomist collects your blood sample on-site.
- The sample is transported to the laboratory.
- Results are uploaded digitally without the need to travel.
Your Results Explained
Low / Negative Levels
No significant dsDNA antibodies detected.
This makes active lupus less likely, though clinical context remains important.
Typical UK reference range:
- Negative: below 10 IU/mL
Borderline Levels
Low-level antibodies detected.
Results should be interpreted alongside symptoms, ANA results and other markers.
Typical UK reference range:
- Equivocal: 10–15 IU/mL
High / Positive Levels
Elevated dsDNA antibodies detected.
This strongly supports a diagnosis of lupus and may indicate increased disease activity, particularly kidney involvement.
Typical UK reference range:
- Positive: above 15 IU/mL
Exact reference ranges may vary by laboratory.

Follow these guidelines for accurate results
Sample Timing
Where possible, attend your blood test between 7am and 12pm. Please wait until any short-term illness or infection has fully resolved before testing. Avoid intense exercise for 24–48 hours beforehand, as this can affect certain markers.
Fasting & Hydration
Fasting is not always required, but some tests may recommend it. If fasting is advised, avoid food for 8–12 hours before your appointment and drink water only. Stay well hydrated, as this helps with sample collection and accuracy.
Medications & Supplements
Continue prescribed medications unless advised otherwise by your clinician. Avoid vitamin, mineral, or biotin supplements for at least 24–48 hours before your test, as these can interfere with results. If you are unsure about any medication or supplement, please let us know before your appointment.
Hormones & Menstrual Cycle
For hormone-related tests, timing within your menstrual cycle may be important. If relevant, follow any specific guidance provided on your test page.
If you have any questions or special circumstances, our team is happy to advise before your appointment.
